Multiple species‐specific molecular markers using nanofluidic array as a tool to detect prey DNA from carnivore scats

چکیده

Large carnivore feeding ecology plays a crucial role for management and conservation predators their prey. One of the keys to this kind research is identify species composition in predator diet, example, prey determination from scat content. DNA-based methods applied detect predators’ scats are viable alternatives traditional macroscopic approaches, showing an increased reliability higher detection rate. Here, we developed molecular method identification wolf (Canis lupus) using multiple species-specific marker loci on cytochrome b gene 18 target species. The final panel consisted 80 assays, with minimum four markers per species, that amplified specifically when high-throughput Nanofluidic array technology (Fluidigm Inc.). As practical DNA collected Sweden. Depending number amplifying required obtain positive call scat, success determining at least one ranged 44% 92%. Although highlight need evaluate optimal sensitive detection, fast cost-efficient tool it also has potential be further other areas large carnivores as well.

Molecular scatology as a tool to study diet: analysis of prey DNA in scats from captive Steller sea lions.

The DNA of prey present in animal scats may provide a valuable source of information for dietary studies. We conducted a captive feeding trial to test whether prey DNA could be reliably detected in scat samples from Steller sea lions (Eumetopias jubatus). Two sea lions were fed a diet of fish (five species) and squid (one species), and DNA was extracted from the soft component of collected scat...

DNA Barcoding: a new tool with wide array of applications

DNA barcoding is a new term introduced in to scientific literatures by Hebert and coworkers almost a decade ago. The concept of barcoding alone is well-known to the public: a series of black bars printed on many commercial products (Universal Product Code), which are used to distinguish different products. Advances made in molecular biology and molecular techniques late 20th century e.g. sequen...

Tigers and their prey: Predicting carnivore densities from prey abundance.

The goal of ecology is to understand interactions that determine the distribution and abundance of organisms. In principle, ecologists should be able to identify a small number of limiting resources for a species of interest, estimate densities of these resources at different locations across the landscape, and then use these estimates to predict the density of the focal species at these locati...

Collembola as alternative prey sustaining spiders in arable ecosystems: prey detection within predators using molecular markers.

Collembola comprise a major source of alternative prey to linyphiid spiders in arable fields, helping to sustain and retain these predators as aphid control agents within the crop. Polymerase chain reaction primers were developed for the amplification, from spider gut samples, of DNA from three of the most abundant species of Collembola in wheat crops in Europe, namely Isotoma anglicana, Lepido...

mac-layer acknowledgment as a tool to detect routing misbehavior

the establishment as well as the survival of mobile ad-hoc networks relies on the cooperation of nodes for performing network operations such as routing and packet forwarding. in these networks, misbehaving nodes can severely degrade network’s performance by not cooperating in networking operations. in this paper, we study the issue of node misbehavior in packet forwarding. to counter this type...
